Cells in the exponential growth phase (mother flask) were collected and resuspended in conditioned (spent) or fresh media. Conditioned media was made by clarifying media from the mother flask. These cell suspensions were then sonicated and used as the diluent to make the working virus solutions (10⁷ PFU/mL)
